After falling into the demon world, Vergil is seen waking up in a pool of blood. In the sky he notices three lights, the eyes of Mundus, which he proceeds to challenge him to a fight. Whatever happened during that fight is unknown, but what became of it was the emergence of Nelo Angelo.
Vergil is presumably seen to have lost his fight against Mundus. As a result he was brainwashed and becames another figure who serves under Mundus' reign. The reason why Mundus did this was probably because of the fact that Vergil is the son of Sparda. Mundus doesn't seem to condemn the fact that Vergil is a son of someone who had betrayed him, but rather he uses it at his disposition. Although Vergil is not a full demon, being a descendent of someone who had the power to stop a war and even defeat Mundus himself is valuable. If not for that, it can be considered a fine way for Mundus to thank Vergil for unlocking and opening the gates to the human world.
One may notice that Nelo Angelo's casual attire is reminiscent to Sparda's attire. That may signify that Vergil may have replaced his father's role and ranking in the demon world. In addition, Nelo Angelo's armor, in particular his helmet, looks similar to Sparda's head shape while he is in Devil Trigger. Also, in this form, Vergil has never been seen in Devil Trigger. In Devil May Cry: Special Edition, Nelo Angelo's Devil Trigger is his armored form. It is a bit illogical to assume that that is his true Devil Trigger because in the first Devil May Cry game Nelo Angelo is seen taking off his helmet. Also, the fact that the suit makes a clanking noise when he walks would seem really odd as a Devil Trigger.

The first time Dante meets his brother as Nelo Angelo was on Mallet Island when he was ordered by Mundus to eliminate his brother. At first, Dante didn't realize that Nelo Angelo was Vergil, although it was hinted to the viewers when Nelo Angelo walks out from Dante's reflection in the mirror. Through the many encounters on the island, Dante complimented Nelo Angelo as someone who had guts and honor. However, was also seen as someone whose talents are wasted for he was working under Mundus. Each time the brothers battled, Nelo Angelo would always win the fights, but it was the little things concerning their pasts which caused him to fail Mundus' orders. After their first fight, as Nelo Angelo had Dante pinned against a wall, his amulet fell from his pocket. Just by a mere glance of it, it caused Nelo Angelo to go berserk. During their second fight, Nelo Angelo removes his helmet to show Dante that he was none other than his brother. Again, Nelo Angelo goes berserk after this fight, and the reason may be because it invoked forgotten past memories because Dante had a flashback after the fight in regards to a past birthday, while he held Vergil's half of the amulet which Nelo Angelo had left behind when he fled. The events were concluded by Mundus telling Trish that Nelo Angelo has been "defeated".
As for the vagueness of the word "defeated", as said by Mundus, what became of Vergil after the events of Mallet Island are unknown. He might have died or he might be still alive.
However, since he is vital to the Devil May Cry series, he is still around, but how will he react to Dante the second time around? Having lost his half of the amulet, and it being what symbolizes his humanity, would Nelo Angelo be more ruthless towards Dante? Would the loss of the amulet be the key to Vergil's acquisition of power? Will Nelo Angelo be in pursuit of retrieving his half of the amulet?
